Drug discovery is a daunting process that requires chemists to sift through millions of chemicals to find a single hit. DNA Encoded library can dramatically speed up the search.
DNA-encoded libraries were first proposed in 1992, in a thought experiment by molecular biologist Sydney Brenner and chemist Richard Lerner, who were then at the Scripps Research Institute in La Jolla, California.
Incubate the active immobilized target protein and DELs for some time; during the time, compounds with strong affinity will bind to the protein while non-bind or weak-bind compounds will be eluted. Heat up to inactive the protein and elute the binding compounds. Through PCR amplification technology, we can read the DNA tags of the binding compounds and then chemical information of the hit compounds can be obtained. After activity assay and chemical structure optimization, lead compounds are generated.
Now the most advanced compound to emerge from GSK’s DNA-encoded library so far is GSK2256294 in PhI (the inhibitor of Pulmonary Disease and Chronic Obstructive). Except GSK,HitGen, in Chengdu, China , is also a member of DEL family and has synthesized more than 6 billion DNA Encoded Compounds.
DNA Encoded Libraries come of age.
