Structure-Data File (SD File) for DMF Submissions

最近2026年1月，收到了FDA要求递交 SD File文件。
We note that there is no SD File in your submission.
The Electronic Submissions Gateway (ESG) has been updated to accept chemical structures as structure-data files (SD File).
Please provide all chemical structures (i.e., drug substance,starting materials, intermediates, and impurities) in a single, comprehensive SD File in 3.2.S.3.2 to facilitate efficient review of your DMF.
The SD file should include the structure of the drug substance, the structure of each other chemical, the name or abbreviation of each chemical as it appears in the DMF, DMF Number, and a unique identifier for cross-reference (e.g., Structure 1, Structure 2,etc.).
The following data items may also be included if available: UNII code2, CAS number, role of chemical (e.g., active ingredient, process impurity, degradant, metabolite, starting material, intermediate).

ESG已更新，可接受化学结构作为结构数据文件（SD 文件）。
请在 3.2.S.3.2 中提供所有化学结构（即原料药、起始物料、中间体和杂质）的单个综合 SD 文件，以方便对您的 DMF 进行高效审查。
SD 文件应包含原料药的结构、其他每种化学物质的结构、DMF 中出现的每种化学物质的名称或缩写、DMF 编号以及用于交叉引用的唯一标识符（例如，结构 1、结构 2 等）。
如果有的话，以下数据项也可包含：UNII 代码、CAS 号、化学物质的作用（例如，活性成分、工艺杂质、降解产物、代谢物、起始物料、中间体）。



虽然法规一直在FDA网站（附件操作说明）上，但是早前一直都没有要求强制提交。
现收到要求，需要提交这个文件，看来后期一般都需要提交了。

通常DMF，一般的eCTD 格式是需要准备的pdf 文件，那现在SD文件是一个sdf的文件格式。


那什么叫什么SD File？
SD File 全称 Structure-Data File ，是一种采用 MOL 连接表格式的化学结构数据文件，可列出一个或多个化学结构。
MOL 连接表格式通过一段文本描述化学结构，其中列出了原子、键、连接性和坐标。
相关数据可以附加到 MOL 连接表中。化学软件将文本转换为化学结构的图像，并将数据放入表格中。
扩展名是.sdf。

啊？3月准备提交一份变更，但是杂质这块没有变化，需要准备上吗？

不翻旧账，以前的都不怎么管。
但是这次的变更，如果有杂质谱变化，可能会被要求，补交。
